Use of stable isotopes to study bioconversion of polyenoic fatty acids in man.

After an overnight fast 4 normal subjects and 4 patients with type I diabetes who had never been given insulin were given dihomo-gamma-linolenic acid 2 g labelled with deuterium, [18, 19-2H4]20:3n-6. Blood was sampled at intervals for up to 72 h thereafter and DELTA5-desaturase activity was studied. It was possible to demonstrate in vivo the conversion of 20:3n-6 to 20:4n-6, as well as the slowness of the conversion, its dependency on insulin and its insufficiency in diabetics.
