Molecular mapping of cocoa chromosomes

A map of cocoa genome has been constructed using isozymes, RAPD, c-DNA and genomic probes. c-DNA library were constructed from germinated seeds of an Amelonado clone.The selection of probes was made on 5 genotypes, using 5 restriction enzymes. 500 tested probes, 33 % of c-DNA and 44 % of genomic revealed polymorphic pattrns. The segregations of markers were studied on a "back-cross type"'mapping population of 100 individuals. After the analises were performed using Mapmaker V.2.0 with a lod score = 3 and 0 = 0.35, it was possible to construct map integrating close to 170 RFLP, RAPD and isozymes markers.
