The development of mycorrhizal infection in coffee seedlings (Coffea arabica L. cv. Catuaí) was studied in conditions of commercial nursery, in substrate with and without organic matter and with different levels of simple superphosphate. The soil utilized was classified as dystrophic Dusky Red Latosol and the source of organic matter used was animal manure (cow's manure). The soil/organic matter employed was 70 and 30 percent (in volume), respectively. The substrate, after the mixture of the two components, was desinfested with methyl bromide. The levels of simple superphosphate utilized were, 0, 1, 2, 3, 4 and 5 kg/m3, and the seedlings were inoculated with the VAM fungus Gigaspora margarita Becker & Hall. One hundred and twenty days after transplanting and inoculation, the plants were harvested. In terms of the growth characteristics evaluated the mycorrhiza increased the dry weight of shoots, stem diameter and leaf area, while the height of coffee seedlings did not show positive response. The mycorrhizal seedlings presented responses of smaller magnitude than the ones reported in the literature. The presence of organic matter in the substrate showed greater responses for all the assessed characteristics, and did not change mycorrhizal colonization of the roots. The effects of P levels of the substrate on the effectiveness of the VAM fungus and colonization of roots, and the possible reasons for the small benefits in the mycorrhizal seedlings are also discussed
