Evaluation of a radial immunodiffusion test for differentiating infected from Rev 1 vaccinated sheep

The usefulness of the Radial Immunodiffusion (RID) test with Brucella melitensis Native Hapten for differentiating infected from Rev 1 vaccinating sheep was studied. It was found that sheep vaccinated with either full or reduced Rev 1 doses were positive in this test and that the post-vaccination evolution of the percentage of positive animals, was similar for both RID and Complement Fixation (CF) tests. When infected animals were studied, CF and RID had similar sensitivity, which was superior to the sensitivity of the Double Gel Diffusion test (DGD) with smooth lipopolysaccharide (S-LPS). Thus, although it is concluded that RID cannot differentiate infected from Rev 1 vaccinated sheep, this test could replace CF for diagnosis
