Standardization of an spectrophotometric technique to measure sperm concentration in the bull, stall ion, boar, ram and dog

The present study was conducted to standardize a spectrophotometric technique for the measurement of sperm concentration in the bull, stallion, boar, ram and dog. Ten ejaculates were obtained from each species from males collected from different sources. A 10-fold dilution of each ejaculate was made,a calibration curve was created and the sperm concentration of each sample was determined and compared by two techniques: haemocytometer and spectrophotometer. The comparison bewteen the two measurement methods showed high correlation with values ranking from 0.92 to 0.99. Using a linear regression equation, a table to calculate a sperm sample concentration was made for each species. Another equipment, RA 50, that also measures sample density was used. The results were compared with the haemocytometer. The correlation between the two tests was good, with values that ranged between (0.90-0.99). The study corroborated the feasibility of using the spectrophotometer or any other density measurement equipment for sperm concentration in several species; it is also a faster method that allows the practitioner or the laboratories to run several samples in a shorter period of time.
