POLYMORPHISMS OF MARKERS ASSOCIATED WITH MILK QUALITY IN CREOLE AND CROSS-BORDER POPULATIONS

Genetic breeding programs have gradually changed from traditional phenotypic selection methods to quantitative and genotypic selection using molecular markers and the identification of genes related to economically important traits. The objective of this work was to identify six single nucleotide polymorphisms associated (SNP) with milk quality genes, DGAT1, CSN1S1, CSN1S2, LALBA, GH1 and ABCG2 in some creole and cross-border populations breeds by sequencing (NGS). Random samples were taken from 73 animals of various pure genotypes, Creoles (Guaymi, GUY and Guabala, GUA), Cross-border breeds (Brahman, BRAH; Holstein, HO; Senepol, SEN; Guaymi, GUY and Guabala, GUA) and cross genotypes (European x Zebu, EXC and Indefinite, SRD). SNP analysis was performed using the Truseq Bovine Parentage sequencing panel. To calculate the genetic variability within population, the following parameters were calculated: Hardy-Weinberg equilibrium, Allelic and genotypic frequency, observed heterozygosis (Hob) and expected (He) and Shannon Index. The markers GH1, LALBA and ABCG2 were monomorphic. The most informative markers were DGAT1, CSN1S1 and CSN1S2, with the DGAT1 marker having the highest Hob and He values with values of 0,424 and 0,430, respectively, and the lowest values for Hob and He were observed in CSN1S2 with 0,247 and 0,276, respectively. These results suggest that the polymorphic markers found may be useful in breeding programs, adding to the quantitative selection, however, further analysis is required, increasing the number of animals and breeds. It was possible to determine polymorphisms in the markers DGAT1, CSN1S1 and CSN1S2, in the genotypes submitted to the present study, however, no alleles fixed in the Holstein and Guabala breeds were observed. The Guabala breed showed a fixation of the A allele in ABCG2, a completely opposite situation in the rest of the genotypes studied.
