Effect of reduced glutathione (GSH) on the sperm quality of verraco under subhumid tropic conditions

In this work was evaluate the effect of reduced glutathione (GSH) on sperm quality of boars under tropic conditions. 40 ejaculates from 4 boars (10 replicates per boar) of commercial lines Yorkshire / Landrace and Pietrain / Duroc between 24 and 28 months of age, were collected at 7 day intervals by the gloved hand technique.The ejaculate was diluted 1:10 with 2000 x106 (viable sperm), L-Reduced Glutathione (GSH, Sigma-Aldrich ®) added to the diluent half conservation (MIII ®). Three groups were formed, control group without GSH (0GSH) and two treatments with GSH concentrations 1 mM (1GSH) and 2 mM (2GSH), total mobility, integrity of the plasma membrane, osmotic resistance and acrosomal integrity initial and in the semen conserved at 17º C was evaluated at 0, 24, 48, 72 and 96 h. The plasma membrane integrity of boar sperm supplemented with 2GSH was 88.0% and 1GSH of 87.3%, different with respect 0GSH (P <0.05). In total mobility, in 2GSH and 1GSH was 58.9% and 56.9%, respectively, showing differences (P <0.05) with respect to the 0GSH (52.0%). During the conservation period from 72 and to 96 h of storage, total mobility showed a 14% decrease for 0GSH, while that in IGSH nad 2GSH only a decrease of 11 and 6% respectively, was observed. Plasma membrane integrity at 96 h was 87.52%, 85.40% and 80.65% for 2GSH, 1GSH and 0GSH, respectively (P<0.05). The variables osmotic tolerance and acrosomal integrity not show significant differences (P> 0.05) in between treatments during the time of conservation. The supplementation with 2 mM of GSH to the dilution medium improvement the quality of spermatozoa of boars maintained in tropical conditions conserved at 17° C

El eyaculado fue diluido 1:10 con 2000 x106 (espermatozoides viables), se utilizó L-Glutatión Reducido (GSH, Sigma-Aldrich®) adicionado al diluente media duración (MIII®). Se formaron 3 grupos, grupo control sin GSH (0GHS) y dos tratamientos con GSH a concentración 1 Mm (1GSH) y 2 mM (2GSH), se evaluó la movilidad total, integridad de la membrana plasmática, resistencia osmótica e integridad acrosomal inicial y en el semen conservado a 17º C a las 0, 24, 48, 72 y 96 h. La integridad de membrana plasmática de los espermatozoides en 2GSH fue 88.0% y en 1GSH de 87.3%, diferentes con respecto a 0GSH (P<0.05). En la movilidad total, en 2GHS y IGHS fue 58.9% y 56.9%, respectivamente, mostrando diferencias (P<0.05) con respecto al 0GSH (52.0%), la integridad acrosomal y la tolerancia osmótica no mostraron diferencias entre tratamientos. O ejaculado foi diluído 1:10 com 2000 x106 (esperma viável), L-Glutationa reduzida (GSH, Sigma-Aldrich®) foi adicionada ao diluente de média duração (MIII®). Foram formados três grupos, um grupo controle sem GSH (0GHS) e dois tratamentos com GSH na concentração de 1 Mm (1GSH) e 2 mM (2GSH), avaliaram-se motilidade total, integridade da membrana plasmática, resistência osmótica e integridade acrossomal inicial .em sêmen armazenado a 17ºC nas horas 0, 24, 48, 72 e 96 h.
