Establishment and protocol optimization to obtaining microsatellites markers in Theobroma cacao using multiplex system in ABI PRISM 377 sequencer
The objective of this work was to establish and to optimize a methodology for obtaining of those markers in cocoa, using the automatic sequencer ABI Prism 377 in multiplex system. The methodology of DNA extraction and PCR reaction is the same as has been used in Cocoa Research Center (CEPEC) for microsatellite in agarose gel. Testing several dilutions, the best procedure was to add 1.5 uL of loading buffer.
